Acetyl-CoA carboxylase-α: Gene structure-function relationships1

چکیده
Acetyl-CoA carboxylase-α (ACC-α) is a key enzyme in the regulation of fatty acid synthesis and is subject to both acute control, via reversible phosphorylation, and chronic control that results in the regulation of synthesis of the enzyme. The gene for ACCα is expressed ubiquitously, but expression is highest in lipogenic tissues: adipose, liver, and lactating mammary gland. These tissues demonstrate a metabolic adaptation to changing physiological demands; for example, during lactation fatty acid synthesis in adipose tissue is markedly repressed, resulting in the partitioning of lipogenic precursors to the mammary gland. Lipogenic tissues can also exhibit dysfunctions that result in excess fat deposition in farm animals and obesity in humans. Transcription of the ACC-α gene is initiated from multiple promoters in a tissue-specific fashion. Promoter II (PII) transcripts are present in all tissues, whereas promoter I (PI) transcripts are principally restricted to adipose tissue. We have also identified an additional promoter (PIII) that is also expressed in a

Genetic Polymorphism Detection of the Exon 1 Region of Acetyl-CoA Carboxylase Alpha Gene in Iranian Mahabadi Goat Breed
Acetyl-coenzyme A carboxylase α (ACC-alpha) is considered as the key regulatory enzyme in fatty acid biosynthesis. ACC-alpha gene is located on Caprine chromosome 11 and is polymorphic in many goat breeds. In the current study, we aimed to find possible single nucleotide polymorphisms (SNPs) in the exon 1 region of the ACC-alpha gene in Iranian Mahabadi goat breed. Genomic DNA was extracted fro...
متن کاملResistance of various biotypes of Canary grass (phalaris. Spp) to acetyl-CoA carboxylase-inhibiting herbicides.
Little seed canary grass (Phalaris minor L.) is a major weed in wheat fields in some parts of Iran. To evaluate the efficacy of molecular and greenhouse methods in detecting the resistance of 49 biotypes of canary grass(Phalaris. Spp) to acetyl-CoA carboxylase-inhibiting herbicides, two methods including whole plant screening and PCR-based molecular methods were applied. Results showed that the...
متن کاملStructure and substrate selectivity of the 750-kDa α6β6 holoenzyme of geranyl-CoA carboxylase.
Geranyl-CoA carboxylase (GCC) is essential for the growth of Pseudomonas organisms with geranic acid as the sole carbon source. GCC has the same domain organization and shares strong sequence conservation with the related biotin-dependent carboxylases 3-methylcrotonyl-CoA carboxylase (MCC) and propionyl-CoA carboxylase (PCC). Here we report the crystal structure of the 750-kDa α6β6 holoenzyme o...
متن کاملArachidonate and medium-chain fatty acids inhibit transcription of the acetyl-CoA carboxylase gene in hepatocytes in culture.
Transcription of acetyl-CoA carboxylase in avian liver is low during starvation or after consumption of a low-carbohydrate, high-fat diet and high during consumption of a high-carbohydrate, low-fat diet. The role of fatty acids or metabolites derived from fatty acids in the nutritional control of acetyl-CoA carboxylase transcription was investigated by determining the effects of long- and mediu...
متن کاملMolecular cloning and characterization of two genes for the biotin carboxylase and carboxyltransferase subunits of acetyl coenzyme A carboxylase in Myxococcus xanthus.
We have cloned a DNA fragment from a genomic library of Myxococcus xanthus using an oligonucleotide probe representing conserved regions of biotin carboxylase subunits of acetyl coenzyme A (acetyl-CoA) carboxylases. The fragment contained two open reading frames (ORF1 and ORF2), designated the accB and accA genes, capable of encoding a 538-amino-acid protein of 58.1 kDa and a 573-amino-acid pro...
